um... every pro airtech has always told me that Teflon tape and air systems dont mix.
if a shred of the teflon tape came loose, and entered the airflow, then it oculd cause the same problem your having now, or more, if it reached the marker regulator.

To back up wat others have said, it will be dirt around the Oring, or a damaged O ring.
Make sure its degassed, take off the fill nipple, clean the oring, check it for damage. replace if you need to. (if you have spares you could just replace it to be safe), then reassemble using locktite on the thread.

NO and NO again, use thread lock, do the above and you will be posting again about your regulator being inconsistant, if the smallest bit of teflon tape gets into the system (it will) then you are in a world of hurt especially if you have a preset reg 'cos to dissasemble them voids the warranty, plus you would probably never even see the piece of tape that is causing the problem. Blue threadlock is the answer use red if you must but be warned it is a much higher strength type (needs more elbow power to remove anything stuck with that type and you could damage the nipple or whatever if you use the red threadlock) so don't say you haven't been warned.

I've used teflon on the nipples of two of my preset tanks without any problems. Thread locker would work just as good though, but as said don't use the red. It takes more than elbow power to get that off, you'll need vise grips, a rubber malet and ultimately a new tank
