This can be done while there still gas in the bottle right as long as the on/off tap is closed right?

Yup. Make ABSOLUTELY sure that the on/off is closed, undo the four bolts on the underside of the manifold, and the reg itself drops right off.
Make sure you either have a diagram, or a good memory for which order everything goes back in though when you undo the springs and poppets and stuff...
